MGS will run client samples on the Qubit® Fluorometer and the Agilent 2100 Bioanalyzer (RNA samples only) as a quality control check before library preparation. The service will also run the client and service prepared libraries on the Qubit® Fluorometer and the Agilent 2100 Bioanalyzer as a quality control check before sequencing on the Illumina MiSeq™ instrument, or before delivery to the OGBF for running on the Illumina HiSeq instrument. Genomic DNA samples are not run on the Agilent 2100 Bioanalyzer because there is no assay to measure DNA >12 Kb in size.
The following Invitrogen® assays are run:
The Quant-iT™ HS DNA Assay or Quant-iT BR DNA Assay is used to quantify genomic DNA samples, PCR amplicons and prepared libraries, and the Quant-iT™ RNA HS Assay is used to quantify total RNA samples. In both instances, the other two assays are run to determine the percentage of contamination. % contamination should be ≤ 20%.
The following Agilent 2100 Bioanalyzer assays are run:
If the samples, client prepared libraries or service prepared libraries do not meet our quality or quantification standards, you will be required to sign an NZGL disclaimer should you wish to continue with the processing of the samples and/or libraries.
