Associate Professor Laryssa Howe

Associate Professor in Infectious Disease/Building Programme Relocation Manager

School of Veterinary Science

Laryssa spent most of her education years in the United States, graduating from Denison University in Ohio with a Bachelor of Science degree in Biology and went on to the University of Pittsburgh to complete her doctorate degree in the Department of Infectious Diseases within the Graduate School of Public Health.

Her studies in Pittsburgh focused on the antibody neutralization properties of a virus called Equine Infectious Anemia Virus (EIAV). She returned to New Zealand in 2004.

As a Senior Lecturer in the Institute of Veterinary, Animal and Biomedical Sciences (IVABS) at Massey University in Palmerston North, Laryssa currently works on developing a better understanding of illnesses related to Apicomplexan parasitic diseases in cattle, sheep and native birds.

Laryssa also lectures on various topics of Microbiology, Immunology and Virology for the veterinary and bachelor of med-lab science programs.

Project Title: Papillomaviruses, p16, and skin cancer in cats

Although papillomaviral (PV) DNA is frequently present in feline cutaneous squamous cell carcinomas (SCCs), a causative association cannot be proven. Oncogenic human PVs cause neoplastic transformation by inhibiting retinoblastoma (pRb) and p53 activity. Therefore, absence of pRb and p53 immunostaining, along with increased p16 immunostaining, indicates a PV etiology in some human SCCs. If PVs cause cutaneous feline SCCs, it was hypothesized that a similar immunohistochemistry profile, along with PV DNA, would be detectable. This was investigated using 5 feline viral plaques, 10 Bowenoid in situ carcinomas, 19 SCCs from UV-exposed skin and 11 SCCs from UV-protected skin. Papillomaviral DNA was amplified by PCR from 30 of 45 lesions. Reduced pRb immunostaining was present in 26 of 45, increased p16 immunostaining in 30, and p53 immunostaining in 19. Both reduced pRb immunostaining and increased p16 immunostaining were more frequent in lesions containing PV DNA. In contrast, no association between p53 immunostaining and the presence of PV DNA was observed. SCCs from UV-protected skin more frequently contained PV DNA, reduced pRb, and increased p16 than UV-exposed SCCs. UV-exposure was not associated with p53 immunostaining within the SCCs. These results suggest feline PVs alter cell regulation by degrading pRb. Unlike oncogenic human PVs, there was no evidence that feline PVs degrade p53. These results provide further evidence that PVs may cause feline cutaneous SCCs, especially those in UV-protected skin, and suggest a possible mechanism of this oncogenic action.

Project Title: Factors influencing pregnancy outcomes in Neopora caninum infection ewes

Neospora caninum is an obligate intracellular parasite and is recognised as the leading cause of bovine abortion worldwide. Natural infection with N. caninum has been described in sheep but it has generally not been regarded as a significant cause of abortion. Recently, there have been several New Zealand cases of foetal abortions where N. caninum was detected which strongly suggested the involvement of Neospora in these abortions. However, there is minimal information about the prevalence of N. caninum infection naturally occurring in New Zealand sheep flocks and particularly its impact on reproduction success. Thus, this present study provides preliminary data on the role that Neospora is playing in ovine reproductive failure by establishing the prevalence of N. caninum antibodies and DNA in ewe blood and foetal material present in 21 New Zealand sheep farms with ongoing unexplained abortion problems and 10 farms with consistently high fertility levels. The results of this study demonstrated an overall seroprevalence of 1.4% which varied between Aborting/non-pregnant (1.8%), age-matched pregnant controls (0.6%) and high fertility (2.1%) ewes. However, despite the variation observed, there was no statistical difference between the three groups. In addition, Neospora DNA was detected by PCR in 13% of submitted foetal brains and in ewe blood from aborting/non-pregnant (6.9%), age-matched pregnant controls (3.6%) and high fertility pregnant (2.1%) ewes. When the PCR results were considered with the IFAT and IDEXX ELISA results, there was no correlation between serology positive and PCR positive blood samples.
